Abstract: Streptomyces malachiticus is a common actinomyce strain in soil. However, there has been little research about the biological activity of its metabolites. This study mainly focuses on screening the enzyme inhibitor of recombined isocitrate lyase (ICL) of Mycobacterium tuberculosis and the enzyme inhibitor of the glyoxalic acid pathway of Mycobacterium tuberculosis as ascreen for anti-TB drugs. Compound 2053-2, dibutyl phthalate (DBP), was isolated from Streptomyces malachiticus metabolites by silica gel column chromatograghy and preparative HPLC. DBP is a potent inhibitor of both ICL and the glyoxalic acid pathway of Mycobacterium smegmatis, which is of significant importance as far as a cure for tuberculosis is concerned.
Key words: Mycobacterium tuberculosis, isocitrate lyase, preparative HPLC, glyoxylate cycle pathway, dibutyl phthalate
