A novel method of probe-melting curve for nucleic aciddetection without occupying fluorescence channel Yang Han;Shangzhi Ji;Xiangzhong Ye;Yimin Li;Yixuan Zhang * 2015 (3):  57-63.  摘要 ( 430 )   A novel method, probe-melting curve method, has been established to increase the detection throughputs within one fluorescence channel. Besides using an asymmetric PCR assay, this method employs another specially designed probe with a 5′ terminus modification. The method has been demonstrated by detection of rotavirus as model system. The sensitivity of probe-melting curve method was proved to be equal to the conventional TaqMan real-time RT-PCR. And the experiment results were cross checked by agarose gel electrophoresis. The successful establishment of probe-melting curve method breaks numbers of fluorescence channels from the restrictions and enlarges the application fields of current real-time PCR instruments. 相关文章 | 计量指标

Digital Identification based on five kinds of enzymeactivity changes for mountain cultivated ginseng withdifferent growth periods Shiyi Xu;Yu Chen *;Lili Zhong;Yu Wang;Jingshuai Wu;Lei Shi;Wei Xu * 2015 (3):  64-68.  摘要 ( 397 )   Panax ginseng has been used as a highly valued medicinal plant. To investigate the biochemical variables of mountain cultivated ginseng roots with different growth periods, five kinds of enzyme activities were measured, respectively. It was found that five enzyme activities were increased remarkably during the slow-growth period relative to the fast-growth period. The activities of AMY and ACP were significantly increased during the whole ginseng growth period. However, EST, ALP, CAT activities were not found changes during the slow-growth period. Enzymatic activities of AMY and ACP can be used as one of the intrinsic indicators to assess the quality of ginseng and identify the growth period of P. ginseng in the future. 相关文章 | 计量指标

Study on the quality control of Shang-Ke Bone-Knitting Tablet (SKBT) Zehai Song;Luhui Li;Qiao Li;Na Han;Zhihui Liu;Jun Yin * 2015 (3):  69-78.  摘要 ( 348 )   Shang-ke Bone-knitting tablet (SKBT) is widely used in China because of its excellent efficiency on bone-fracture healing, which is composed of 12 kinds of drugs. In order to improve and strengthen the quality control standards of SKBT, three drugs, Asterias Amurensis (hai xing), Olibanum (zhi ru xiang), Myrrha (zhi mo yao), were chosen because of their contributions to the efficacy of SKBT and were identified by thin-layer chromatography (TLC). The results showed that the TLC method we have developed was reliable, accurate, simple and easily operated. The method was appropriate to control the quality of SKBT. Except for the TLC part, a simple, rapid and sensitive high-performance liquid chromatography (HPLC)-UV method has been developed to simultaneously quantify three active saponins of Notoginseng Radix et Rhizoma (notoginsenoside R1, ginsenoside Rg1 and ginsenoside Rb1). Methodological investigation indicated that the developed HPLC assay could be readily utilized as a quality control and quantitative analysis method of SKBT. 相关文章 | 计量指标
