Abstract: The petal of Rosa rugosa Thunb. was widely used in Xinjiang as traditional uygur’s medicine in treating liver-depressed hypochondrium pain, perturbed, anxiety, bad memory, gastric cavity bloated and tuberculosis etc. Three major bioactive components with similar polarity in this plant were first successfully purified by high-speed counter-current chromatography. More importantly, they were proved to be significant behavior for hepatoprotective, strong antioxidant and anti-inflammatory.The separation procedure was performed in two steps with two solvent systems. Under the condition of system (1): n-hexane-ethyl acetate-methanol-water (2:5:1:5, v/v), 1.5 ml/min for flow-rate and preferable revolution speed of 900 rpm, fraction I and two compounds of avicularin (10 mg) and juglanin (9 mg) were purified from 100 mg of crude extract. Fraction I , got from system (1), was isolated by the system (2): ethyl acetate-methanol-water (20:1:20, v/v). Four mg of astragalin was got from the fraction I. The compounds of avicularin, astragalin and juglanin were got with the purity of 98.88 %, 98.06 % and 95.03 %, respectively, as determined by HPLC. The structures of compounds were identified by HPLC with reference substances and further confirmed by UV and NMR spectrum. This method was able to purify the three flavonoids at high purity and yield levels, starting from a very complicated sample.
